Dependence of Prolonged Water Absorption by in Vitro Locust Rectum on Ion Transport

چکیده

1. An in vitro preparation of the locust rectum, consisting of an everted cannulated sac, is described. After the first hour, the rates of ion and water transport, the electropotential difference and tissue volume do not change significantly over the next 4 h, i.e. a steady-state situation is approximated. 2. The activities and properties of these rectal sacs are reasonably similar to those in vivo; they produce hyposmotic absorbate, exhibit rectification of osmotic flow, and absorb water from pure sucrose solution for 1 h. Inhibition of absorption by KCN + IAA and also ouabain indicates metabolic dependence. 3. Analysis of absorbate shows that tissue ions are secreted to the haemocoel side when water is absorbed over the first hour from pure sucrose solutions. Prolonged transport of water (i.e. from second to fifth hour) requires the presence of ions on the lumen but not the haemocoel side. This is the first direct evidence that water absorption in the insect rectum is coupled to ion transport as previously postulated. The location of the proposed ion recycling suggested by these observations is discussed.

K⁺ absorption by locust gut and inhibition of ileal K⁺ and water transport by FGLamide allatostatins.

The scanning ion-selective electrode technique (SIET) was utilized for the first time in Locusta migratoria to characterize K(+) transport along the digestive tract and to determine the effect of two locust FGLamide allatostatins (FGLa/ASTs) on K(+) transport: a previously sequenced FGLa/AST from Schistocerca gregaria (Scg-AST-6; ARPYSFGL-NH2) and a newly sequenced FGLa/AST from L. migratoria (...

Transport Properties of Locust Ileum in Vitro: Effects of Cyclic Amp

1. Short-circuited locust ilea exhibited electrical properties remarkably similar to those of recta. The large short-circuit current (Isc) and transepithelial potential, both indicating net active absorption of anions, initially began to decline to zero but could be fully restored by adding cyclic AMP or extracts of corpus cardiacum and ventral abdominal ganglia to the haemocoel side. 2. Ion su...

KC1 transport across an insect epithelium: characterization of K-stimulated Cl absorption and active K transport.

The kinetics of (36)C1 fluxes across cAMP-stimulated, short-circuited locust rectum were studied. Raising external K(+) from 0 to 100 mM increased both K(t) and V(max) for net Cl transport (J(CI)net) by four- to six-fold. Hill plots of J(CI)net indicated non-cooperative Cl interactions. The sequence for cation stimulation of J(CI)net was K > Rb > Cs > Na > NH4 . Low levels of K were stimulatory...

Investigation of ion transport and water content properties in anion exchange membranes based on polysulfone for solid alkaline fuel cell application

In present research work, homogeneous anion exchange membranes based on polysulfone (QAPSFs) were prepared via chloromethylation, amination and alkalization. In amination step, trimethylamine and N,N,N',N'-tetramethyl-1,6-hexanediamine were used as amination and crosslinking agents, respectively. The chloromethylated polysulfone was characterized by 1HNMR spectroscopy and chloromethylation degr...

Actions of Ion-transport Peptide from Locust Corpus Cardiacum on Several Htndgut Transport Processes

1. Schistocerca gregaria ion-transport peptide (Scg-ITP), a neuropeptide isolated from locust corpora cardiaca, stimulates ileal Cl~ transport (/«:) in a dose-dependent manner and causes increases in Na, K (/K) and fluid reabsorption (7V) as previously observed with crude extracts of corpus cardiacum and with cyclic AMP. Unlike cyclic AMP, ScgITP does not stimulate ileal NH4 secretion. 2. H sec...
